Preptin Deficiency Does Not Protect against High‐Fat Diet‐Induced Metabolic Dysfunction or Bone Loss in Mice

چکیده

Preptin is derived from the cleavage of E-peptide pro-insulin-like growth factor (IGF)-II and an insulin secretagogue. Observational studies have linked elevated circulating preptin to metabolic dysfunction in humans; however, a causal role for has not been established. Additionally, can promote osteoblast proliferation differentiation, suggesting link with skeletal health. We previously described global knockout (KO) model. In this study, we sought uncover impact KO mice on response moderately high-fat diet (HFD) low-fat (LFD). HFD groups had higher weight fat mass gain, lower trabecular cortical bone volume fracture load, liver triglycerides. males, deficiency led blood glucose than wild-type (WT) under LFD conditions. This was accompanied by differences microarchitecture, including fraction, number, thickness. These were absent female mice, although females HFD-driven increase triglycerides that WT mice. Female increased glucose-stimulated secretion conditions Overall, may detrimental metabolism positive health male protect against storage while enabling islet compensation When consider serum levels are humans both sexes pathological states which elevated, comorbidity risk needs be better understood. © 2023 The Authors. Protease-activated receptor-4 deficiency does not protect against bleomycin-induced pulmonary fibrosis in mice.

Protease-activated receptor (PAR)-4 is a thrombin receptor expressed in lung tissue. Previous in vitro data showed that PAR-4 triggers epithelial-to-mesenchymal transition, which is crucial in pulmonary fibrosis. These observations prompted us to investigate the role of PAR-4 in a murine model of bleomycin induced pulmonary fibrosis. We observed that wildtype and PAR-4 deficient mice developed ...

Tumor Progression Locus 2 (Tpl2) Deficiency Does Not Protect against Obesity-Induced Metabolic Disease

Obesity is associated with a state of chronic low grade inflammation that plays an important role in the development of insulin resistance. Tumor progression locus 2 (Tpl2) is a serine/threonine mitogen activated protein kinase kinase kinase (MAP3K) involved in regulating responses to specific inflammatory stimuli. Here we have used mice lacking Tpl2 to examine its role in obesity-associated in...

Pnpla3/Adiponutrin deficiency in mice does not contribute to fatty liver disease or metabolic syndrome.

PNPLA3 (adiponutrin, calcium-independent phospholipase A(2) epsilon [iPLA(2)ε]) is an adipose-enriched, nutritionally regulated protein that belongs to the patatin-like phospholipase domain containing (PNPLA) family of lipid metabolizing proteins. Genetic variations in the human PNPLA3 gene (i.e., the rs738409 I148M allele) has been strongly and repeatedly associated with fatty liver disease. A...

Calcium does not protect against colorectal neoplasia.

Calcium could decrease risk of colorectal neoplasia by binding bowel-irritating compounds and diminishing mucosal proliferation. This study quantitatively summarizes epidemiologic studies addressing this hypothesis and aims to explain heterogeneity between studies. Twenty-four articles reported 43 measures of relative risks (RRs). The weighted mean, according to a random effects model, did not ...

Knowledge does not protect against illusory truth.

In daily life, we frequently encounter false claims in the form of consumer advertisements, political propaganda, and rumors. Repetition may be one way that insidious misconceptions, such as the belief that vitamin C prevents the common cold, enter our knowledge base. Research on the illusory truth effect demonstrates that repeated statements are easier to process, and subsequently perceived to...
